fluorescent cytotoxicity assay [View Printable]

I need to do a cytotoxicity assay with NK cells, tumoral cells and an antibody I have producted, but using fluorescence instead of the usual Cr 51. I want to try the calcein, but I know that a lot of other probes exist. Does anyone use it or an other one, and what do you think is the best? I also would like to know, what are the parameters the most important to perfect such a protocol? Thank you everybody.

There are tons of different cytotoxicity assays out there. The best assay really depends on your application. I've used the MTS assay, sulforhodamine B assay and also I've recently tried an ATP-based luminescent assay. You can measure different things with the various assays (membrane integrity, mitochondrial enzyme activity, LDH release, etc), so the the mechanism of cytotoxicity may dictate exactly sort of assay you use. Also, you might be limited by the filters available for your plate reader (if you're doing a plate based assay). Remember to include a lethal control - I like staurosporine the best, but it's expensive.
