Are there any other users of the Ciphergen mass spec system? I would like to exchange tips and protocols on obtaining high quality results.

I have heard that IDD(Institute for Drug Development) in San Antonio, Texas is doing a great job: their laboratories are equipped with a mass spectrometry-based Ciphergen Protein Biology System. The analytical method utilizes a proprietary ProteinChip technology for resolving and identification of proteins in microliter size samples of complex biological mixtures. I never worked with them but you might be able to contact them and get some information: IDD Phone: 210-677-3800 FAX: 210-677-0058 www.ctrc.cc

I heard that their MS is not very good. However, they've come up with Seldi chip compatible interfaces to other MS made by other vendors.

Ciphergen is running two User Meetings in May this year. User meetings are being held on May 1 4th in Newport, Rhode Island and May 9 -10th in Berlin, Germany. These meeting give you a real opportunity to meet other Ciphergen customers and I would urge you to try and attend one of them. Also, have you tried calling your local Ciphergen field scientist and asking for the names of some other customers in your area (Geographical or interest)?

We have a 4.5 year old Ciphergen PBS2 and use it daily. Depending on the types of results you want or expect, it is either a fairly decent MS or not up to the task. We have used it for profiling KO animals with amazing results, following treatment of animals with various drugs with good results. We have also done C-terminal sequencing with carboxypeptidase Y. Others in our group have used it to map fragments from limited tryptic digests for structural analysis. Do you have specific questions or needs? As for their ms not being very good.... that is a matter of opinion. What people either forget or don't know is that after the chips are washed and put in the machine, the process is strictly maldi-tof ms. Is it high end? no, is it reproducible, yes. Can other machines use the chips, yes,,, but from the very limited experience i have had on a maldi-tof/tof using the ciphergen chips, i would say that that is not the best arrangement as the chip surface is very rough compared to the maldi-tof/tof's own target.
