I am setting up experiments that involve phosphorylated and total ERK calculations.
I heard about colorimetric and chemiluminescent immunometric products that are available.
Can any one recommends a company with a product they have used?

Dear Sandy,

I can think of two companies: 1- A company called : Active Motif, Inc. at www.activemotif.com with a product called FACE or Fast Activated Cell-based ELISA to analyse protein phosphorylation.
2- A technology developed at DiscoveRx commercialized under the name HitHunter HTS assays for GPCRs and Kinases.
using chemiluminescent signals.

The ERK ELISA kit from Active Motif, Inc is fine. Give a try.

if you are interested in doing quantitative immunoblotting there are some good antibodies to regular and phospho-erk...i cannot remember now exactly who they were from since it was a while ago that i used them, but i am pretty sure that one of them was from santa cruz.

i know it's been a while since you posted this inquiry, but i just noticed today and ad on the back of the scientist for their cell signaling assays. if you buy one and use it and tell them the results they send another one free (though the issue is from 2/28 so the offer might not still be on). anyway, one of the listed kits is ELISA based total and phosphospecific erk.

Sigma does a great phospho-ERK1/2 antibody (WB 1:1000) always worked for me. Use with ECL+ reagents (Amersham) for quantitative western blotting if you have a STORM imager for scanning blots directly by chemifluorescent detection (image is saved directly and can be quantified using associated IMAGEQUANT software). Santa cruz ERK2 antibody is good for total ERK expression (WB 1:2000)

I regularly use immunoblot and quantitatively take IDV from the chemilimunisence developed hyperfilms or from Gel-doc software itself.
