I am working on insilico ligand design against certain cancer antigens. I have a lead compound structure obtained through wet lab resutls (mass spec data).

I wanted to know about informatics software (open source/free ware) that will generate large sets of compounds automatically, by retaining the backbone structure of my compound but will modify the various side chain residues. (more like a lead compound library)

can you suggest any?
