Numerous Universities now have facilities for the production of transgenic animals and knockouts. I had some generated at UCSD http://cancer.ucsd.edu/Research/Shared/tgm/default2.asp
I was both pleased with the price and the results.

Knockout mice contain the same, artificially introduced mutation in the gene of interest in every cell, abolishing the activity of a preselected gene.

RNAi (RNA interference) refers to the introduction of homologous double stranded RNA (dsRNA) to specifically target a gene's product, resulting in null or hypomorphic phenotypes. The difference is that the introduction of RNAi into a cell, targets the messenger RNA for the protein of interest, rather than mutating the gene to shut down its expression.

Here is a company that can make custom-made gene overexpression, humanized and Knock-down (RNAi) mice models in 2 months. We haven't used them but I thought I would pass on the info.
