We are currently looking to identify a biotinylated protein by MS. The biotin we used cross-links with the side chains of Lysine residues.
Nobody I have spoken to seems to know whether biotinylation of Lysines will affect the subsequent trypsinization (which cleaves after lysine residues) of the protein. If it does, then the fingerprint we will obtain will be difficult to interpret, if not, then it should be okay.
Anybody have any experience with MS and biotinylated proteins?

Thanks Guy,
Its not exactly what I was looking for but I have emailed the group that performed this work as I have a feeling that they will have a specific answer to my question.
All the best.
