Hi,
I am trying to find some information about the vector pONG. Many ages ago, we received some constructs (sans map) that were in this vector. Now it has fallen to me to work on them, and I have been unable to find out a shred of info about this vector.

Does anyone know anything about it, or (gasp!) have a map???

YIKES! Been there...all my sympathies, brother!

When receiving questionable constructs from unreliable sources, I always defer to the credo of my dear mentor:

1) Trust no one.
2) Believe nothing.
3) Be bitter.

What is it you need to know? What do you want to do with these constructs? I'm going to assume you want to ultimately express the proteins in cells, or perhaps merely subclone portions of the inserts...

Even if you found a vector map, you wouldn't necessarily be able to trust that someone didn't somehow modify the vector.

Do you at least know the cloning sites? If so, I would cut out the inserts and put them into a vector I could trust.

Otherwise, send them for sequencing with all avaliable the typical primers (T3, T7, SP6, M13 forward, M13 reverse, anything else you have on hand) and hope that a short read will give you cloning information at both ends...then again, put them into a vector you can trust.

Unless there is something VERY special about this vector, start over! THEN DRAW A MAP!!

Please let the board know how you solve this problem...we really care!!
