Hi all,

Could anybody tell and guide me to the best protocol for cell lysis (At the end my needs are nuclear proteins). I have quite a number of protocols with me but confused which one would be best for protein extraction. I am working on adherent cells and need a protocol that will serve my purpose.

Vdas please read this article: Andrews, N.C. and Faller, D.V. (1991) A rapid micropreparation technique for extraction of DNA-binding proteins from limiting numbers of mammalian cells. Nucleic Acids Research 19, 2499

Hi all,

is the protocol described for extraction of DNA-binding proteins good for Mass spec studies  also ??

I am interersted in whole protein extraction instead of organelle proteins. I am trying various methods of protein extraction.

Please suggest which lysis buffer and method one would  be the best ??

 The article of Andrews & Faller of 1991 clearly answers the question. For mass spec studies I am not sure because both have different protocols.
