I am using E. coli TG1 strain. To select for the F sex pilus I have to streak it on minimal medium plates. I understand that the minimal medium is based on M9 salts, MgSO4, CaCl2 and glucose or other carbon source [Sambrook et al.] but I do not know if I have to supplement with amminoacids/vitamins. If anyone has a recipe for minimal medium to grow specifically TG1 E. coli please let me have it or tell me where I can search for it. Thanks.

how long does it take to grow TG1 E. coli strain on M9 minimal plates? I streaked them yesterday and no colony showed up after overnight incubation at 37 °C. Do I have to wait longer times?

Yes, it does take longer for cells to come up - you may have to wait about 30-40 h before it even comes close to a LB after o/n incubation. M9 minimal is as per Sambrook - however, a net search reveals many other recipes (don't know about TG1). Remember that the glucose/other carbon source has to be autoclaved separately from the salts, in a different time/temp combination. Supplements are usually not required, but one tip is to add ~10% of a sterile tissue culture medium such as DMEM without any additives to your autoclaved medium.
