I am Mayur.


I am new in this field . plaese help if u know the following


CAn lysis buffer contaiing ( Mgcl2,Nacl,leupeptin,aprotinin,banzamidine,Na. pyrophosphate, EGTA calcium chloride ) be used for protein estimation by BCA followed by liquid scintillation counting. normaly cell lysis by 0.1 Naoh is used ?

some of the mentioned substances like EGTA interfere with BCA assay. you have o reduce the concentration of these components to a level where they donot interfere with estimation. Also the proteases that you are adding are proteins so they will add to total protein estimation error.

With scintillation counting, the buffer is compatible. But use hydrophilic or hydrophobic scintillation cocktail depending upon if your protein is hydrophilic or hydrophobic.
