Did anyone do western blot of cytosolic / mitochondrial cytochrome C release? I transferred the protein in semi-dry blotter for 40 minutes and the band was very weak (from ECL detection and exposed for 10 minutes). Could anyone give me some advice? Thank you first!

Thanks to your suggestion first. Fix with paraformaldehyde may be help, but if it compatible to staining solution? As I need to fix the cell and then stain the nucleus and cytoplasm. THX!

I think may be you can run a PCR and then agarose gel first to see any signal before taking it to real-time PCR. Do you use SYBR green or TaqMan? TaqMan is accompanied with exogenous control.

Hi Samm, Thanks for your advice first. My lab also used this method to see apoptosis, but it is a rather qualitative method than a quantitative one. We use PI to see cell cycle and annexin-V-FITC to show early and late apoptosis. I see paper that some people use 7AAD to quantify apoptosis, and ...
