Posted comment on blog   Who invented the computer?

The easy answer would be 'Alan Turing'. Obviously several other people made significant contribution from developing the idea- using electronics for calculation to actually building such a machine. I found this site to be nice. http://www.turing.org.uk/turing/scrapbook/computer.html There are sever ...

Posted new topic in Model Organisms forum   large particleflow cytometry for C. elegans, D. melanogaster

Most C. elegans labs do large scale screens for mutants, gene upregulation, downregulation or developmental effects (smaller or longer body. Most of these were done under a microscope by an individual in the lab. The size of worms- about 1mM for the adults- makes them too big for cell sorters common ...

Thanks. I'd try the PEG next time. (I usually wash my beads with RIPA buffer *3 wash with PBS resuspend the beads in 2X Laemmli buffer Boiling and blotting the usual way.) No, there is no verloading. There is extensive backgroung only when I use anti-mouse-HRP. I am planning to switch to ...

Hi all. I am trying to pull down a transcription factor from C. elegans lysate. I can pull down the TF with a rabbit GFP ab and blot it with a GFP-HRP conjugate. I can pull down almost all of the TF. I do not get any background bands or smear. I am looking for a interaction between this transcrip ...

Replied to topic in Microbiology forum   Silencing kinases in mammalian cells large screen

[quote=R Bishop]The answer is they didnt validate them all. "The RNAi strategy employed was validated by targeting 53 randomly chosen genes with 3 siRNAs each and performing quantitative real-time PCR (qRT-PCR) analysis to determine the level of knock-down achieved in each case. For 13 of these g ...
