Oligo DT for cDNA synthesis

Hi Folks,


I am using Oligo DT from Promega to prepare cDNA from RNA. They provide it in 20 ug quantity. I ound out its too expensive and its very small quantity. I use like 1 ul for every 20 ul reaction. Am I using too much or its really expensive?


I was looking to find some alternative, i.e. Oligo DT from some other company like Thermo Scientific. Looking forward to hear from you people. Please help me out if you know anything abt it.

Some people in our lab also use Promega's oligo dT (product C1101) which costs £47 for 20 micrograms.


The protocols require about 0.5 micrograms per cDNA synthesis so it's just over £1/reaction.

Hi Umesh,


I guess it depends on the oligo(dT) primer you are using. For the classical one (12-18 nucleotides) using 0.5 ug is more than enough to convert micrograms of RNA into cDNA. If you are using less RNA you can always lower the mount of primer, but my recommendation is not to cut costs at this step of the process since the later steps (qPCR, etc) tend to be a lot more expensive. Half a microgram of oligo(dT) primer costs about $1 -$2.
