curcumin isolation

curcumin isolation

I want to isolate curcumin from the commercial crude curcuminoid mixture. However, the poor solubility of curcumin makes it difficult to purify by column chromatography. What are the best conditions? Should I just load my column with a very large volume of solution????

Extraction and isolation of curcuminoids from the methods section:
"turmeric powder was successively extracted with 95% ethanol at room temperature, then precipitated with petroleum ether to yield crude curcumin mixture consisted of 78% curcumin I, 16% curcumin II, and 5% curcumin III. The crude mixture was then fractionated by silica gel 60 column chromatography using first CHCl3 and then CHCl3/methanol with increasing polarity to isolate pure fractions of curcumins I, II, and III. The identity and purity of each curcuminoids were verified using by TLC, HPLC, IR, MS, and NMR analysis."

      I had a similar query.. not much experience in this field, I wanted to isolate flavonoids, n had a reference to use the chloroform/ methanol gradient system in column chromatography to elute various flavonoid fractions, however, i need a detailed explaination from somebody experienced in it, as to what solvent be used to make the column, since if i use chloroform, it disturbs the bed formation, also, what volume of each gradient be used for elution. pls help out.

                 I am having the same query as yours. Its been 4 months since you posted the query. so am asking you the  same.. Will you be able to guide me on the solvent used to pack the column for isolating curcumin? I used water to pack the column. but it allows poor or no penetration of the sample and eluent (chloroform/methanol). ple help me.

Turmeric Oleoresin 100 gms is mixed with 100 gms of E.D.C. AND STIR AND KEEP IT FOR ONE WEEK AND THEN SUBJECT THE MIXTURE IN NUTCH FILTER ,CRYSTALISED CURUCUMIN GETS SEPEARTED AND STAYS IN FILTER.
REMOVE THE PASTE AND DRY IT IN DRYER TO GET CURUCUMIN.

 I want to get min 70%

 I want to get min 70% efficiency in isolation of curcumin from oleoresin by using Ethyl Acetate as a solvent.Please help me .Im getting only 60 % efficiency. How  can i improve it.
