CROSS-PALMGRENS METHOD

SECTIONS

8 micron paraffin wax sections

SOLUTIONS

1. Reducer

Pyrogallol 10g

Distilled water 450ml

Absolute alcohol 550ml

1% Nitric acid 2ml

2. 1% ammonia

3. Silver Solution

Silver nitrate 15g

Potassium nitrate 10g

Distilled water 100ml

5% Acetic acid 1ml

4. 0.2% Gold Chloride

5. 5% Sodium Thiosulphate

METHOD

Place reducer in 60C incubator for use at stage 5
Take sections to water
Place in 1% ammonia for 5 minutes
Rinse in distilled water
Impregnate with silver solution for 30-45 minutes
Drain excess silver solution from the slides and without rinsing, pour on the warm reducer until the sections turn brown. Slight agitation prevents deposit formation
Wash in distilled water

Tone in 0.2% gold chloride for 2 minutes

Wash in water

Fix in 5% sodium thiosulphate solution

Wash in water

Dehydrate, clear . Mount sections in DPX
