I have amplified 5 kb gene cluster (uaing Acc LA taq polymerase with proofreading, Sigma) responsible for surfactant production. Primer was designed with EcoRI and HindII restriction site, can I ligate my restricted fragment with pGEMT easy vector?

 Hola yes you can, in this way you will have a source of your insert for subcloning in an expression vector, because cutting with EcoRI and Hind II you will recover your fragment that you could isolate in a agarose gel selecting the 5Kb band .
