colony pcr for gram positives ?

is there a method to do colony pcr for gram positive isolates? if yes, i need the procedure.

We do colony PCR on gram positives, Group A and Group B Streptococcus. We pick a single colony into 10 ul of water in a PCR tube and microwave it for 2 minutes. Then we use 1 ul of this as a template in a PCR. You don't want to use any more than 1 ul since there are some inhibitors. Also you need to prepare this fresh right before setting up your PCR and can't reuse it on a different day. Hope this helps!
