Sorter FACSAria

Hi everyone..

I´ve just started to use a FACSAria II and I´ve some questions, hope you can help me..

Can I acquire samples with some culture medium if they are phenol free? They can´t have ir any serum right?

The cell suspentions have so be in PBS or FacsFlow or they can be, for example, in salt water?

I think next days I´ll have much more questions..

The cell suspensions should indeed be phenol red free as this can sometimes interfere with the light refractivity of your sample.  I'm not sure about the serum issue however.

 I think as long as you avoid

 I think as long as you avoid the phenol red and too much protein you will be OK.  High levels of protein (serum) can clog up the system.  I usually use something like 0.3% BSA.

 I know people use different

 I know people use different salt solution (PBS, HBSS etc) and I believe I've seen mention of people sorting marine organisms with salt water.  You probably have to switch the sheath fluid to filtered salt water as well as I'm concerned that if the osmolarity of the sample and sheath fluid is too different that you could have problems.  I haven't dealt with this issue, so I can't give you a certain answer.  Maybe someone else can on this forum, or you can google or give BD a call for advice.
