I want to remove the glycosylation from the proteins inorder to check if the different isoforms of glycoprotein will merge into the same MW product after the removal of glycosylation.
I dont have PNGase and cant purchase that either.... Can any chemical induce cleavage achieve the same results???
To mention more The protein of my interst is feebly expressed and its almost 2-3ng content in the 100 microgram protein load as observed by densitometry of tricine resolved gels
Argerine,
Trifluoromethanesulphonic acid (TFMS) treatment of the protein will result in complete removal of all glycosylations and doesn't destroy the peptide bonds. Very informative paper here.
Cheers,
Hi Suola
thanks for the quick reply and the informative paper.
Do you work on the glycoproteins???
