Hi, I performed a PCR and did a gel electrophoresis with a positive and negative sample, and a DNA sample, but got no bands. What could be the cause of this? (lanes 4-6) Lane 8 is the HILO marker.

Hi arfernan,

Hi arfernan,

The reason for a PCR reaction to fail are many, from bad DNA, primers that do not work, PCR conditions that are not ideal, etc.

My recommendation is to get a set of primers you know works, and a control DNA you know is of good quality, and run those side by side with this PCR that is not working. If the control DNA, with the control primers, work, then you can assume the problem is with your primers. In that case it may be a good idea to design new primers, or optimize your PCR conditions to see if you can get these primers to work.

Good luck
