High molecular weight genomic DNA was isolated using DNA extraction buffer as described previously (Zarlenga et al., 1991). After incubation at 37 C for 30 min, RNA was digested by adding DNase-free RNase A (10 µg/ml). PCR was performed with Taq DNA polymerase (Perkin-Elmer, Foster City, California) with the use of 3 random primers, A-03 5'-AGT CAG CCA C-3', A-08 5'-GTG ACG TAG G-3', and A-20 5'-GTT GCG ATC C-3' (OPA; Operon, San Diego, California) in a Thermal Cycler 480 (Perkin-Elmer). The cycling parameter consisted of 45 cycles of 94 C for 1 min, 37 C for 1 min, and 72 C for 2 min.
