Accumulation of membrane protein in cytoplasm

The membrane protein I am studying is accumulating in the cytoplasm. How do I determine why this is happening?

hello matrix, you can isolate

hello matrix, you can isolate these membrane protein and identify them using MS. may be  there is  any change in aminoacid residue  because of that they are accumulating in cytoplasm. not only that you can also check PTM in these proteins as well as in those protein whc are localized in the membrane and can find out is there any relation between them. gud luck

 Is this occuring only under certain circumstances, i.e. when the cell is stressed?
Also what organism are you working with?

I am working with human cells

I am working with human cells. The protein is found in the cell membrane normally. When the cell becomes neoplastic, high levels of the protein is found in the cytoplasm. We have knocked that protein down. We now want to transfect the cell with the same protein, and target it to the membrane and compare biological consequences.
