Selection of Calcium sensitive probes based on Kd

Hi,

Can anyone explain to me how to select a calcium sensitive probe for a study. For example Fura 2 and Fura FF. One is low affinity dye and the other high affinity. How does the Kd values of these dyes help to chose which dye is best suited for study.

Basically Fura FF has a much lower affinity for Ca2+ than Fura-2 and so can be used when you expect to record calcium signals in concentrations >1 µM  and for recording fast Ca2+ transients with high fidelity.
Fura-FF is also poorly sensitive to magnesium (Mg2+) and so tends to suffer from is less interference than a dye like mag-Fura-2.
