I am trying to find information about the promoter region for a few human microRNAs, miR-124-1, miR-124-2, and miR-124-3. Ultimately, I want to make a luciferase promoter construct for each of these miRNA genes with the most appropriate upstream length (I've heard less than 2 kb is enough?). When I try to use some common promoter web-based software like Cold Spring Harbor Laboratory's Mammalian Promoter Database, they don't seem to like miRNAs and don't recognize their accession numbers.
Does anyone have any suggestions on how to find these miRNA promoters, and what is an appropriate length to clone into the plasmid? Also, I'm pretty new at this, so I'll welcome any advice or tricks to making and using these constructs.
