what buffer should I use with DNA ???

my experiment is study the transition of DNA  ; from B-DNA to Z-DNA and I have to adjust the pH to physiological pH (7.4) . But i don't know what buffer shoud I use ? I hesitate between Tris-buffer or Phosphate buffer , but I have to add the metal ions in the solution,so I think the phosphate buffer is not the good choice because it will precipitate with the metal ions. Please suggest me... Thank you for your kindness. Sorry if my english is bad...

Hi, Jangjy.

Hi, Jangjy.

As per my experience you should go for tris  buffer, because DNA is stable  in same buffer. Most of the kit provide same buffer for  DNA storage .

I agree with the response you

I agree with the response you received (choosing Tris over Phosphate). Although the amount of salt will not be significant during pH adjustment of the Phosphate buffer, DNA is 'happier' in Tris-based buffer.
I would be aware of the buffers that come with the kits though, since some of them are at pH 8 or 8.5
