i am working with human fibroblasts and want to arrest them in prophase. metaphase is too late! Does anyone have any ideas??
This is a tough one, mainly because it is so early in the cell cycle. There is an old reference to MST-16, which can arrest cells at prophase, but in all honesty I do not have experience with this. Alternative you can use IBMX (3-isobutyl-1-methylxanthine), but again I do not have experience with this.
Perhaps you can use siRNA to knockdown RAD51C? Knockouts of this gene arrest in prophase.
A person in my old lab used to work on CDK (Cyclin dependent kinase) inhibitors and I think that these inhibitors can induce cell cycle arrest in prophase.
