Two patients, A and B, have their serum alkaline phosphatase level measured.
Patient A has an enzyme specific activity which converts 8 µmoles of 4-nitrophenol phosphate substrate/min/100 ml blood and has a 60% haemocrit (this indicates that 60% of the blood volume is made up of cells).
Patient B: 0.1 ml of serum was used (haemocrit also 60%) and this produced 14 µg of 4-nitrophenol product (molecular weight 139) in 30 minutes under the standard assay conditions.
