Hello to all,

Is there any body got the idea about total estimation of a.a?

I have been trying to find out the method for estimating total amino acids from tendons or collagens.

This method is required to normalize the hydroxyproline concentration to total aminoacids of the tendons or collagens instead of normalizing to dry or wet weight of the mouse tendons [b]. Normalizing the hydroxyproline con. to tendons weight incorporate the errors, because mouse tendons weight only few mgs

Actually i am interested in total protein estimation (i don't know it is correct terminology) from acid hydrolyzed protein samples. I got an paper published in analytical biochemistry292, 125-129 (2001), in which the author described the Ninhydrin based method for the quantification of total protein.

Presently i am using this method to quantify total protein (even though ninhydrin doesn't react with proline) from acid hydrolyzed collagen sample and reading against hydrolyzed BSA protein.

Usualy what I am doing is to

Usualy what I am doing is to an astimation after running the DNA on an Agarose gel.
Running it in paralel to a 1kB ladder and doing it corresponding to it.
It is writen how many ng per each marking of the ladder per each step.
