Conjugating antibodies

Hi, I've been let down by Santa Cruz, who are now unable to deliver the mAb they sold me (HLA-DQ, clone TAL 4.1). Unfortunately it's only available in unconjugated form from any other suppliers, so I guess my only option is to conjugate it to PE myself. My PI is keen for me to use a kit, I was just wondering if anyone could recommend an easy to use kit which has given them good results?

Any help you can give would be gratefully received!

Hi! I've used the Pierce EZ-Label kit for FITC with good results, and they now have both regular and microscale labelling kits for a whole range of flurophores corresponding to the ALexa family of dyes at multiple wavelengths (including one for PE) - I think its called daylight or dyelight or something of the sort.

Molecular Probes (Invitrogen)

Molecular Probes (Invitrogen) also sells somes great reagents with a wide variety of Alexa-tags. They have worked well for me in the past. The kit is EASY!

The Dylight Kits from Pierce

The Dylight Kits from Pierce worked well for me. They are the fastest kits I've seen (took me lkess than an hour) and my results were clean. Here's the link:

Hello,
have you, has somebody you know, tested DyLight Kits with other NHS-activated dyes?
And a second question, do the coupled antibodies still recognize the epitopes?
I have problems with other coupling kits and want to switch to a working solution, perhaps it could be the PIERCE Kit!

Thank for your posts everyone - just to report back that I used the Invitrogen Alexa-fluor 558 kit and it seems to have worked very well. The coupled antibodies are binding well to my cell lines. Thanks again!
