Antimicrobial Screening in Organs and Tissue of Snakehead Fish.

 hello sir and mdm,

i am currently doing by B.Sc Biotechnology.in my final year research i m encountering some problems.my topic is Antimicrobial Screening inOrgans and Tissues of Snakehead Fish (Channa Striatus}. I followed  Kirby-Bauer Technique using few organs such as kidney ,liver,blood,meats,gills,scale,skin,ovary, and intestine.but the result were obtained which no inhibition in all the organs using acidic extraction.
An organic solvent in acidic condition was used in the preparation of crude extracts.

so can i know any possible reasons why there is no inhibition zone in alll the organs?

Maybe you could try broth

Maybe you could try broth-microdilution o 2 layer radial diffusion on agar.As i understand that your agar have no zone of inhibition..either your sample has no activity or your sample doesn't work well on agar.....maybe you could try using your whole sample without adding any solvent.directly diffuse on your disc.....gud luck!!!

i m going to test the extracts of internal organs n tissues of snakehead fish against human pathogens such as ,Acinetobacter baumannii,Escherichia coli,Klebsiella pneumoniae,Micrococcus luteus,Salmonella enteritidis and another few bacterias.so that i can identify any antimicrobial property within an internal organs of the fish that can inhibit the growth of bacteria..
