O mv Holding potential

 Hello, I've a big question, I've been working with GABA activated currets the last months and my advisor suggested me to keep my cells in 0 mv holding potential since we didn't have TTX to block sodium currents and our recordings could be contaminated with sodium noise if we hold the cells at -60 or -40 mV... my question is if it is valid to to work with this holding potential since it's not a physiological condition.

If you are trying to isolate

If you are trying to isolate the GABA currents whatever method you use to block or inactivate the other channels expressed in the cell is not going to be "physiological".

If you are expressly trying the characterize the biophysics / pharmacology of the GABA channels then you just need to be confident that you are studying a pure GABA signal.
