I have a question in data analysis. I like to measure the peak value of K channel current. I do not apply leak-subtraction metnod (P/N).
When I apply the holding potential (-80 mV), the current line always shows about -50 to -100 pA. I believe this is a leak, since this current value X membrane resistance is close to the holding potential. Also, this exsits after application of K channel inhibiter.
Should I measure the peak current from 0 (zero) line, or should I measure the current from the base line (the current at the holding potential)? Maybe small impact, but I like to understand.
