Cardiomyocyte patch clamping

Hi,
I am working on adult rat cardiomyocytes. When trying to bring the patch pipette near the cardiomyocytes to form giga ohm seal they start contracting. Any suggestions?

You must use/increase EGTA in the pipette solution till 5, 10 or even 20mM, because the contractions are due to free calcium inside the cell. Search papers in the pubmed from Fischeister, R, (France) because this author works a lot with cardiomyocytes.

I think you meant

I think you meant Fischmeister R.
Click the link below to get the details of the pubmed search

It finds 45 references

 Hey

 Hey

I'm also working on rat cardiomyocytes using patchclamp to measure Calcium current. I have a problem isolating viable cells. Which protocol do you use? How good is your cell condition?

Thanks
