I am working with Axopatch 1D, Clampex-9 on HEK293T cell lines, recently we had problem of incorrect capacitance displayed in the software, now with the technical assistance from axon that problem is solved. But I would like to calculate capacitance experimentally, So can anyone please suggest a good reference describing the protocol and a simple method followed to calculate Memebrane capacitance of a cell.

But I am looking for some protocol in voltage clamp or current clamp mode to be used with my setup to measure the capacitance
Usually I use the value of C-slow as membrane capacitance.
If you like to obtain the experimentally measured value of capacitance, I would record an electrotonic potential in order to obtain the value of taw. From that, I would be able to obtain the value of the membrane capacitance. The following is the method.
With current clamp mode, I would pass a small (negaive) current, and you would obtain the electrotonic potential (from the data, you can measure the membrane resistance and taw).
From the equation of:
taw = R x C
You would obtain the value of C with your experimental data. This should be the valueof the capacitance of your cell.
