Can anybody tell me about there observation using K252a a kinases inhibitor, works as RTK inhibitor at 200nM , and not blocking Mapk activation.

You have to be careful. ERK can be activated not only by Trk receptors (blocked by K252a) but also by calcium entry. So, we have to be sure if you don't have calcium entry in your cells that would activate ERK.
So, it means if a system which allows Ca++ entry will not be compatible for using K252a as a blocker of Trk Receptor, what else can one use to check the receptor Trk B involment?
and does anybody have observed that MAPK is blocked using K252a and at which conc.?
