Protocol for: Released ROS by Using BSA H2HFF
Abstract or Description:
Our research group is studying the signaling pathway involved in the release of pro-inflammatory cytokines from macrophages; an important step in this process is the regulation of the redox balance within the cell. Using the Carestream Image Station 4000MM system (IS4000MM), we have been able to visualize the production of reactive oxygen species mediated by the activation of NADPH oxidase by macrophages stimulated with extracellular ATP. Changes in fluorescence were imaged using the IS4000MM using a 470 nm excitation filter and a 535 nm Wide Angle (WA) emission filter. Images captured using this system allow us to characterize the involvement of redox signaling in macrophages based on the amount of fluorescent intensity measured in the region of interest (ROI).
